Parkinson’s disease (PD) is phenotypically characterized by the loss of dopaminergic neurons in the substancia nigra leading to the clinical symptoms of tremor and rigidity. Many recently identified genes involved in PD are related to mitochondrial dysfunction and energy metabolism. To understand how genetic risk factors manifest into the clinical picture, one has thus to follow the effect of mutations from the molecule to the cell and the tissue. We address this challenge by an interdisciplinary approach that combines multiscale imaging methods and metabolomics with mechanistic spatiotemporal modeling to investigate the role of brain energy metabolism in neurodegeneration at different levels.
