Ultima 2P+ Two-Photon microscope 

The Ultima 2P+ system is an upright, cutting-edge two-photon microscope renowned for utilising a gentler approach to fluorescence excitation. Employing two-photon illumination, this system excites fluorophores using longer, lower-energy wavelengths, resulting in superior resolution while minimising photodamage. This technique proves particularly effective for imaging a diverse range of thick and dense, light-scattering samples, making the Ultima 2P+ a powerful tool in biological research. 

High-Sensitivity deep tissue imaging   

  • Studying the intricate details of brain structures and functions deep within tissue.  
  • Monitoring the progression of diseases or the effects of treatments in deep-seated organs.  
  • Observing cellular and molecular processes in thick tissues, such as tumor environments or dense connective tissues.  

Photo-stimulation 

  • Manipulating neuron activity in neuroscience research, specifically in understanding neural circuits and behaviour through techniques like optogenetics.

Largest Field of View and Superior Field Uniformity 

  • Comprehensive imaging of large tissue sections or whole organisms, providing a broad context for biological studies.  
  • Detailed, uniform observation of developmental processes in embryos or organoids over time, ensuring consistent image quality across the entire sample.  
  • Large-scale mapping of neural networks or vascular systems, facilitating the understanding of complex systems with high resolution and clarity across extensive areas.  
  • 2Photon Illumination is a gentler way of exciting fluorophores – it reduces photobleaching and is also less phototoxic to the sample, therefore enabling long-term imaging 
  • 2Photon Illumination facilitates penetration deeper into thick and/or dense samples 
  • 2Photon Illumination provides increased spatial resolution when compared to standard excitation techniques – the chance of 2 photons hitting the same fluorophore at precisely the same time is low so most of your fluorophores will not be emitting and will not be contributing to the haze from out-of-focus planes  
  • Galvo scanning field of view: 22.7mm Field Number with80% uniformity (1.11mm x 1.11mm with Nikon 16x 0.8 NA objective)  
  • Over scan galvo field of view: 28mm Field Number (1.4mm x1.4mm with Nikon 16x 0.8 NA objective) 
  • X-Y translation base for Ultima 2Pplus and Investigator Plus systems. Includes motors and translation optics. 
  • 8kHz resonant galvanometer 
  • High-speed shutter to protect PMTs from light pulses.  
  • ET-DSRed (TRITC/CY3) filter set in cube for fluorescence viewing via the eyepiece.  
  • Filters ET545/30x and ET620/60m; dichroicT570LP. Not required for 2P imaging. 
  • High speed switching box for control of galvo and Pockels cell signals.  
  • For use with all 2Pplus, Investigator Plus, and mONP nosepieces with small dovetail attachment. Allows for high speed z-positioning of objective lens. 400 um range. Capacitance feedback algorithm used with positioning accuracy of 25nm.Includes Piezo stage, controller and an M32-M25 slim objective adapter. Other adapters available for purchase.
